預(yù)期用途
PuriMagTM WCX富集試劑盒（基于弱陽(yáng)離子交換色譜法）是為在 MALDI-TOF 質(zhì)譜分析之前基于離子交換相互作用從生物樣品中富集蛋白質(zhì)和肽而開發(fā)的。
產(chǎn)品信息
PuriMagTM WCX：10 mg/ml×1ml，存儲(chǔ)于 20%乙醇溶液中。 產(chǎn)品應(yīng)小心儲(chǔ)存和處理，以避免污染。
產(chǎn)品在常溫下運(yùn)輸。我們建議到貨后將試劑盒存放在 2 - 8°C 環(huán)境中。請(qǐng)勿冷凍！
WCX 緩沖液
Binding buffer (BB): 0.05 M Ammonium Acetate (AmAc), pH 4;
Washing buffer (WB): water (HPLC grade);
Elution buffer (EB)：1% TFA in water;
推薦的生物標(biāo)志物富集程序
通常，我們推薦以下樣品制備方案，但根據(jù)樣品的種類和應(yīng)用，可能需要調(diào)整此方案。為此，可以改變不同的參數(shù)（例如，使用的磁珠量、樣品和結(jié)合溶液的體積）。
結(jié)合樣品到磁珠
1. 將磁珠在渦旋裝置上充分混合 1 分鐘。
2. 將10 μl BB 和 20-40 μl 的WCX 磁珠轉(zhuǎn)移到標(biāo)準(zhǔn)薄壁 PCR 管中，并上下吹打混合。

3. 向溶液中加入5μl血清，上下移液5次，充分混合。

4. 孵育 5 分鐘。

5. 將試管放入磁力架上，將磁珠收集在試管壁上1分鐘。上清液應(yīng)澄清。

6. 使用移液管小心移除上清液。避免吸頭與磁珠接觸，并注意不要取出磁珠。
洗滌結(jié)合到磁珠上的樣品
7. 向試管中加入100μl的WB。

8. 在磁力架中來(lái)回移動(dòng)管子十次，注意磁珠的運(yùn)動(dòng)。
9. 將磁珠收集在管壁上 1 分鐘。
10. 使用移液管小心地去除上清液。
11. 重復(fù)步驟 7-10 兩次。
從磁珠上的洗脫樣品
12. 加入5μl的EB，通過上下集中移液10次來(lái)分散管壁上的磁珠。

13. 將磁珠收集在管壁上 2 分鐘。

14. 將透明的上清液轉(zhuǎn)移到新鮮的試管中。
15. 繼續(xù)執(zhí)行如下所述的適當(dāng)MALDI-TOF靶標(biāo)制備方案或適當(dāng)方法儲(chǔ)存樣品。
